The defender, 24, is a target for Wolves, but Tony Pulis’ side are offering more money than Mick McCarthy’s club are.
Reading are reportedly willing to let their captain go for £3.5 million, an amount which Stoke are prepared to meet.
Wolves are understood to have already had a bid in June of £3m turned down by the club.
Stoke made the decision to move for the former England under-19 international when they were put off highly rated centre-half Scott Dann by Birmingham City’s £12m valuation.
The St Andrew’s defender is understood to be Pulis’s first-choice but he has changed his primary transfer target to Mills.
The Royals’ centre-back has played at numerous teams, including Manchester City, Southampton, Colchester and Doncaster Rovers before signing for Reading in 2009 for an undisclosed fee believed to be in the region of £2m, but the club are now willing to let one of their prize assets go after failing to win promotion again last season.
The club reached the play-off final but lost 4-2 against Swansea City, with Mills scoring a header.
